Abstract Biomass is a versatile renewable energy source that can be used in all parts of the energy system, but it is a limited resource and usage needs prioritisation. Here we use a sector-coupled European energy system model to explore the range of cost-effective near-optimal solutions for achieving stringent emissions targets. We show that provision of biogenic carbon rather than energy is the main value of biomass, with the energy system cost increasing by 20% if biomass is excluded. It is not crucial in which sector biomass is used if it is combined with carbon capture to enable negative emissions and e-fuel production. A shortage of renewable electricity or hydrogen primarily increases the value of biomass for fuel production, which appears as the marginal abatement option and is most sensitive to uncertainties. Biomass usage is significantly affected if the biomass is associated with upstream emissions.

Abstract Energy Demand Reduction (EDR) refers to lowering the amount of energy required to provide energy services across mobility, shelter, nutrition or the production of goods and services, among others, with the goal of reducing greenhouse gas emissions. In recent years, global studies have attempted to understand the contribution EDR could make to climate mitigation efforts. Whilst these studies are important to build a global picture, climate targets and policy are necessarily devised at the national level. To address this disconnect, we develop a bottom-up, whole system framework that comprehensively estimates the potential for energy demand reduction at a country level. Replicable for other countries, our framework is applied to the case of the UK where we find that reductions in energy demand of 52% by 2050 compared with 2020 levels are possible without compromising on citizens’ quality of life. This translates to annual energy demands of 40GJ per person, compared to the current OECD average of 55GJ. Our findings show that EDR can reduce reliance on high-risk carbon dioxide removals, moderate investment requirements, and allow space for ratcheting up climate ambition. We conclude that national climate policy should increasingly develop and integrate EDR measures to both articulate national ambition and feeding into international pledges through Nationally Determined Contributions.

In this report, Azithromycin (Azi) antibiotic was measured by carbon paste electrode (CPE) improved by graphene nanoribbon-CoFe 2 O 4 @NiO nanocomposite and 1-hexyl-3 methylimidazolium hexafluorophosphate (HMIM PF 6 ) as an ionic liquid binder. The electrochemical behavior of Azi on the graphene nanoribbon-CoFe 2 O 4 @NiO/HMIM PF 6 /CPE is investigated by voltammetric methods, and the results showed that the modifiers improve the conductivity and electrochemical activity of the CPE. According to obtained data, the electrochemical behavior of Azi is related to pH. under optimum conditions, the sensor has linear ranges from 10 µM to 2 mM with a LOD of 0.66 µM. The effect of scan rate and chronoamperometry were studied, which showed that the Azi electro-oxidation is diffusion controlled with the diffusion coefficient of 9.22 × 10 -6 cm 2 /s. The reproducibility (3.15%), repeatability (2.5%), selectivity, and stability (for 30 days) tests were investigated, which results were acceptable. The actual sample analysis confirmed that the proposed sensor is an appropriate electrochemical tool for Azi determination in urine and Azi capsule.

The introduced work represents an implementation of the automatic benchtop electrochemical station (BES) as an effective tool for the possibilities of high-throughput preparation of modified sensor/biosensors, speeding up the development of the analytical method, and automation of the analytical procedure for the determination of paracetamol (PAR) and dopamine (DOP) as target analytes. Within the preparation of gold nanoparticles modified screen-printed carbon electrode (AuNPs-SPCE) by electrodeposition, the deposition potential E DEP , the deposition time t DEP , and the concentration of HAuCl 4 were optimized and their influence was monitored on 1 mM [Ru(NH 3 ) 6 ] 3+/2+ redox probe and 50 μM DOP. The morphology of the AuNPs-SPCE prepared at various modification conditions was observed by SEM. The analytical performance of the AuNPs-SPCE prepared at different modification conditions was evaluated by a construction of the calibration curves of DOP and PAR. SPCE and AuNPs-SPCE at modification condition providing the best sensitivity to PAR and DOP, were successfully used to determine PAR and DOP in tap water by "spike-recovery" approach. The BES yields better reproducibility of the preparation of AuNPs-SPCE (RSD = 3.0%) in comparison with the case when AuNPs-SPCE was prepared manually by highly skilled laboratory operator (RSD = 7.0%).

The ability to detect low numbers of microbial cells in food and clinical samples is highly valuable but remains a challenge. Here we present a detection system (called 'APC-Cas') that can detect very low numbers of a bacterial pathogen without isolation, using a three-stage amplification to generate powerful fluorescence signals. APC-Cas involves a combination of nucleic acid-based allosteric probes and CRISPR-Cas13a components. It can selectively and sensitively quantify Salmonella Enteritidis cells (from 1 to 10 5 CFU) in various types of samples such as milk, showing similar or higher sensitivity and accuracy compared with conventional real-time PCR. Furthermore, APC-Cas can identify low numbers of S. Enteritidis cells in mouse serum, distinguishing mice with early- and late-stage infection from uninfected mice. Our method may have potential clinical applications for early diagnosis of pathogens.

The importance of gut microbiota in human health and pathophysiology is undisputable. Despite the abundance of metagenomics data, the functional dynamics of gut microbiota in human health and disease remain elusive. Urolithin A (UroA), a major microbial metabolite derived from polyphenolics of berries and pomegranate fruits displays anti-inflammatory, anti-oxidative, and anti-ageing activities. Here, we show that UroA and its potent synthetic analogue (UAS03) significantly enhance gut barrier function and inhibit unwarranted inflammation. We demonstrate that UroA and UAS03 exert their barrier functions through activation of aryl hydrocarbon receptor (AhR)- nuclear factor erythroid 2-related factor 2 (Nrf2)-dependent pathways to upregulate epithelial tight junction proteins. Importantly, treatment with these compounds attenuated colitis in pre-clinical models by remedying barrier dysfunction in addition to anti-inflammatory activities. Cumulatively, the results highlight how microbial metabolites provide two-pronged beneficial activities at gut epithelium by enhancing barrier functions and reducing inflammation to protect from colonic diseases.

Since the 1950s, Staphylococcus carnosus is used as a starter culture for sausage fermentation where it contributes to food safety, flavor, and a controlled fermentation process. The long experience with S. carnosus has shown that it is a harmless and "food grade" species. This was confirmed by the genome sequence of S. carnosus TM300 that lacks genes involved in pathogenicity. Since the development of a cloning system in TM300, numerous genes have been cloned, expressed, and characterized and in particular, virulence genes that could be functionally validated in this non-pathogenic strain. A secretion system was developed for production and secretion of industrially important proteins and later modified to also enable display of heterologous proteins on the surface. The display system has been employed for various purposes, such as development of live bacterial delivery vehicles as well as microbial biocatalysts or bioadsorbents for potential environmental or biosensor applications. Recently, this surface display system has been utilized for display of peptide and protein libraries for profiling of protease substrates and for generation of various affinity proteins, e.g., Affibody molecules and scFv antibodies. In addition, by display of fragmented antigen-encoding genes, the surface expression system has been successfully used for epitope mapping of antibodies. Reviews on specific applications of S. carnosus have been published earlier, but here we provide a more extensive overview, covering a broad range of areas from food fermentation to sophisticated methods for protein-based drug discovery, which are all based on S. carnosus.

A method using an ammonia electrode is being developed for investigating the deamination of amino acids and amides by bacteria. Application of this method to Campylobacter jejuni and C. coli has led to the demonstration of D-asparaginase activity in some strains. This has allowed the subdivision of both species into D-asparaginase-positive and -negative biotypes. Even though the method is in the developmental stage, it was found to be generally reproducible and easy to perform. Areas for further improving the procedure have been identified. The ammonia electrode offers the theoretical possibility of investigating the breakdown of any amino acid by bacteria. It thus opens up a new and practical approach for separating species and strains, particularly in those bacterial groups that are difficult to subdivide by conventional means.

Woody biomass is highly recalcitrant to enzymatic sugar release and often requires significant size reduction and severe pretreatments to achieve economically viable sugar yields in biological production of sustainable fuels and chemicals. However, because mechanical size reduction of woody biomass can consume significant amounts of energy, it is desirable to minimize size reduction and instead pretreat larger wood chips prior to biological conversion. To date, however, most laboratory research has been performed on materials that are significantly smaller than applicable in a commercial setting. As a result, there is a limited understanding of the effects that larger biomass particle size has on the effectiveness of steam explosion pretreatment and subsequent enzymatic hydrolysis of wood chips.

The high incidence of breast cancer is the greastest threat to women' health all over the world. Among them, HER-2 positive breast cancer has the characteristics of high malignancy, easy recurrence and metastasis, and poor prognosis. Traditional Chinese medicine (TCM) has a rich theoretical basis and clinical application for breast cancer. TCM believes that blood stasis syndrome is one of the important pathogenesis of breast formation and development. Taohong Siwu Decoction (TSHWD) is based on the "First Prescription of Gynecology" Siwu Decoction. It is widely used in various blood stasis and blood deficiency syndromes, mainly in gynecological blood stasis. Clinical studies have found that THSWD can treat breast cancer by reducing blood vessel and lymphangiogenesis with auxiliary chemotherapy. In this study, we aim to explore the material basis and mechanism of THSWD in the treatment of HER-2 positive breast cancer through literature review and network pharmacology studies. Through a literature review of the traditional application, chemical composition of Chinese herbal medicine of THSWD, as well as its clinical reports and pharmacological research on breast cancer treatment. Meanwhile, we conducted "component-pathway-target" network through network pharmacology reveals the main material basis, possible targets and pathways of THSWD in inhibiting HER-2 positive breast cancer. Literature review and network pharmacology research results had predicted that, baicalein, kaempferol, caffeic acid, amygdalin, quercetin, ferulic acid, gallic acid, catalpol, hydroxysafflor yellow A, paeoniflorin in THSWD are the main effective chemical composition. THSWD regulates 386 protein targets and 166 pathways related to breast cancer. The molecular mechanism is mainly to improve the microenvironment of tumor cells, regulate the process of tumor cell EMT, and inhibit tumor cell proliferation and metastasis. This study revealed the mechanism of action of THSWD in the treatment of HER-2 positive breast cancer through literature review and network pharmacology studies, providing a scientific basis for clinical application.
